How to Read a
Peptide COA

A Certificate of Analysis can provide useful information—but a professional-looking PDF does not automatically prove that a peptide is legitimate, accurately dosed, sterile, or safe.

A polished report can still leave important gaps. Read each category independently and notice what the laboratory did not test.

Step 1

Match the sample to the product

The report should clearly identify the sample it describes. The lot number on the COA should match the lot printed on the vial. A test from an unrelated or older batch tells you very little about the product in your hand.

  • Peptide or compound name
  • Vial strength or sample description
  • Lot or batch number
  • Testing and report dates
  • Vendor or submitting customer, when disclosed
  • Sealed production vial versus loose sample
A passing result from one submitted vial is evidence about that sample. It does not prove that every vial in the batch is identical.
Step 2

Confirm the peptide’s identity

Identity testing asks whether the sample actually contains the claimed compound. This is commonly evaluated with mass spectrometry: the measured molecular mass should align with the expected molecular mass of the peptide.

A purity result alone does not necessarily confirm identity. A sample can contain one highly pure substance and still be the wrong substance.

Step 3

Understand HPLC purity

HPLC purity is commonly expressed as the percentage of the total detected chromatographic peak area attributed to the primary component. It does not necessarily mean that the peptide makes up that percentage of everything physically present in the vial.

That percentage does not automatically mean:

  • The vial contains 99.2% of the labeled milligrams
  • The vial is correctly filled
  • The sample is sterile
  • The product is free of endotoxins
  • The peptide remained stable in transit
  • Every substance in the vial was detected
Step 4

Find the quantitative peptide content

Purity and peptide quantity are separate measurements. Look for a result labeled peptide content, net peptide content, assay, or quantitative peptide mass, and confirm which testing method was used.

If the report specifically identifies 11.4 mg as measured peptide content, the submitted vial tested above its labeled strength. A result of 8.7 mg would indicate that the submitted vial tested below its labeled strength.

Do not assume total vial mass equals active peptide content. Dried material may also contain water, salts, counterions, or excipients.

When using a reconstitution calculator, a credible quantitative result from a batch-matched report may provide a more accurate basis than the labeled vial strength. Testing one vial still does not establish that every vial contains exactly the same amount.

Step 5

Check which safety-related tests were performed

HPLC purity and identity testing do not establish sterility or injectable safety. Separate tests answer separate questions. If a result is not listed, do not assume the test was performed.

  • Sterility
  • Bacterial endotoxins
  • Bioburden
  • Residual solvents
  • Heavy metals
  • Other specified contaminants
Even a reported purity of 99% does not mean a product is sterile or safe to inject.
Step 6

Check the dates

Review the date received, date tested, report date, and lot production date when available. A report from years ago—or one repeatedly used for newly sold batches—should not be treated as proof of current batch quality.

Step 7

Verify the laboratory and report

Whenever possible, verify the report directly through the laboratory’s website or by contacting the laboratory. Do not rely solely on a QR code that leads back to the vendor’s own website.

  • Laboratory name and address
  • Contact information
  • Report or sample number
  • Testing methodology
  • Authorized signature or approval
  • Independent verification link or QR code
An authentic laboratory report can still be unrelated to the vial being sold. Verification and batch matching are both necessary.

Common COA red flags

None of these automatically proves fabrication, but each warrants closer scrutiny.

The vial and COA lot numbers do not match
The report does not name the compound
Only a purity percentage is shown
Identity testing is missing
No quantitative peptide content is provided
The same COA is used for every batch
The laboratory cannot verify the report
Dates or sample details appear altered
Charts are cropped or analytical data is missing
Repeated near-perfect results lack supporting detail
“Pass” appears without the result or method
Purity is presented as proof of sterility
A loose sample was tested instead of a sealed vial

The bottom line

A useful peptide COA should help answer four separate questions:

Is it the correct compound?
How pure is it?
What quantity was measured?
Were sterility and endotoxins tested separately?

Many vendor COAs answer only the first two—and some answer only one. A COA is one piece of evidence, not an automatic guarantee. Match the batch, verify the laboratory, read the actual results, and pay close attention to what was not tested.
